I am sharing a pAbmAbs generated literature review of KO validated antibodies. Here citing Mao et al. International Journal of Biological Sciences, 2021.
Mao et al. validate this antibody for detection of endogenous p130 Cas/BRAC1 by Western blotting of H1975 and H1299 cells. Clear bands were observed in both H1975 BCAR1-KO and H1299 BCAR1-KO, but the band intensity did decrease compared to the control, along with this Mao et al. also showed a Western blot of A549 cells which overexpressed BCAR1 showing an increase in band intensity (figure 2A).
Based on the available data, we have assigned a three-star rating because of the band intensity originating from an incomplete KO, non-specific binding of the antibody or similar, furthermore no supplementary figure was attached to clarify this, but we highly encourage other scientists to share their observations.
