Review of: Serine/threonine protein kinase PINK 1, Cell Signaling Technology, 6946 by pAbmAbs Data Mining
Our team has carried out deep research in the published scientific literature to identify papers and reports using antibodies directed against human Serine/threonine-protein kinase PINK1 and followed up by extraction and evaluation of the available data to assess its performance according to pAbmAbs rating criteria.
YCharOS is here conducting a large head-to-head study comparing 13 commercial research antibodies against human Serine/threonine-protein kinase PINK1 encoded by PINK1.
The collection of antibodies was tested using HAP1 WT and PINK1 KO cells in the application WB (WT and KO cell lysates/30 µg protein) according to YCharOS standard protocols (see reference). Cells were treated with 10 µM of CCCP (Carbonyl cyanide 3- chlorophenylhydrazone) from MilliporeSigma (cat. number C2759) for 24h.
In WB (1:1000 ab dilution) 6946 detected a band of appr. 63 kDa in WT cell lysate (CCCP positive) with no corresponding band observed in KO cell lysate. Faint bands of higher and lower molecular weight were observed in both genotypes.

I am sharing a pAbmAbs generated literature review of KO validated antibodies. Here citing Miyahara et al. Scientific reports.
Miyahara et al. validate this antibody for detection of endogenous human PINK1 by Western blotting of human breast cancer cells (MCF7), to demonstrate the biological consequence of removing PINK1 (i.e., change in BRCA1 behavior). No band was observed in PINK1-KO. (figure 1h).
Based on the available data, we have assigned a five-star rating, because of strong signal, clean background, but we highly encourage other scientists to share their observations.
