We found two peer-reviewed publications specifically describing the validation of APC-120 from Alomone Labs.
In Johnston et al., J Physiol, 2008, APC-120 was used for Western blotting using brain tissue from CBA mice, detecting a clear band of the expected size of 110 kDa. No band was observed following pre-incubation with blocking peptide or using brain tissue from homozygous Kv2.2 knockout mice (Lexicon, TF1551) (Figure 2B).
APC-120 detected Kv2.2 immunoreactivity in MNTB principal neurons, with prominent labelling in a putative axon initial-segment region. The localization was supported indirectly by electrophysiological evidence for functional Kv2-containing delayed-rectifier channels.
Kihira et al., JBC, 2010 replicated the specificity of APC-120 for Western blotting using homogenates of rat brain neurons as samples by pre-incubation with blocking peptide, completely blocking the signal (Figure 2A). APC-120 also specifically detects rat Kv2.2 but not Kv2.1 in transfected HEK293 cells (Figure 2B).
APC-120 immunostaining of rat brain was compared with staining obtained using the independent mouse monoclonal Kv2.2 antibody K37, which targets a different epitope, supporting the specificity of APC-120 for Kv2.2 in immunohistochemical applications (Figure 5B).
