Review of: G protein-activated inward rectifier potassium channel 2, Alomone Labs, APC-006 by pAbmAbs Data Mining
We found one peer-reviewed literature report specifically describing the validation of APC-006 from Alomone Labs.
In Fernandez de Velasco et al. Scientific Reports 2017 the authors demonstrate the specificity of APC-006 in Western blotting (WB) and immunostaining (IF).
Adult mouse hippocampal lysates and neonatal hippocampal neuronal culture lysates from wild-type and Girk2 knockout animals were Western blotted with the Alomone GIRK2 antibody (Fig 2B). The GIRK2 band is present in WT and absent in knockout material, supporting specificity for WB in this context. The authors further show that APC-006 specifically recognizes recombinant mouse GIRK2 but not rat GIRK1 or GIRK3 in WB using lysates from transfected HEK293 cells (Supplementary Fig. 1).
Furthermore, cultured hippocampal neurons from WT and Girk2 knockout mice were stained using the same Alomone GIRK2 antibody. Specific neuronal staining seen in WT is lost in knockout cells, supporting IF specificity (Fig. 2C-D).
We have rated the antibody based on the available information and according to the pAbmAbs review criteria.
We highly encourage other scientists to share their experience.
