Our team has carried out deep research in the published scientific literature to identify papers and reports using APC-001 and followed up by extraction and validation of the available data to assess its performance according to pAbmAbs rating criteria.
APC-001 detects ROMK1 reliably in WB and IF. In Liu et al., JASN 2015, the antibody showed a high-MW ROMK1 band abolished by siRNA and absent in KO kidneys (Fig. 1), and IF staining reduced by siRNA (Fig. 4). Earlier studies confirmed specificity in IF and WB via antigen pre-adsorption and cross-validation with a second antibody (Mennitt et al. 2001, Fig. 1 and Fig. 5) and reported the expected ~45 kDa band in kidney medulla (Gimelreich et al. 2001, Fig. 10). Recent human biopsy work showed correct collecting duct localization (Sung et al. 2021, Fig. 3), though without negative controls. Overall, strong multi-modal validation supports a 4★ rating: robust for WB, solid for IF, but not yet 5★ due to heterogeneous banding and limited KO-based tissue IHC.
