We found one paper that explicitly validates AAC-001 entitled Flow fluorometry quantification of anion channel VRAC subunit LRRC8A at the membrane of living U937 cells by Yurinskaya et al., published in Channels, 2020
The application was live-cell flow cytometry / flow fluorometry in U937 cells
The authors state they validated the use of flow cytometry with the LRRC8A antibody, and found endogenous cell-surface LRRC8A could be quantified in living cells. They compared unstained cells, secondary-only controls, and primary+secondary staining; whole-cell P1 signal was higher than autofluorescence and secondary-only controls (Figure 1). They also conclude microscopy fluorometry was not sensitive enough under the same antibody concentration.
Dilution: 3 ul in 50 ul cell suspension.
