What do reviewers generally think about sc-9042 from Santa Cruz Biotechnology?

Researchers report poor specificity for the β2 adrenergic receptor in western blotting, with multiple bands between 25 and 125 kDa and no expected 65 kDa band in CHO cells expressing β1, β2, or β3 receptors. One reviewer points out that immunohistochemistry of murine ureter and stomach gave distinct staining at 1:50, but western blotting at 1:400 of CHO cells stably expressing β2 showed no signal difference versus non-expressing lysates, leading to a conclusion of no target specificity.

Hamdani & van der Velden, VU University Medical Center Amsterdam, tested this antibody using CHO cells expressing human beta adrenoreceptor subtype 1, 2 or 3.

In WB, the antibody recognized several protein bands between 25 and 125 kDa in all three cell lines, but no band of the expected size of 65 kDa was observed.

The specificity of this antibody was evaluated in immunohistochemistry of murine ureteres and stomach tissue, as well as western blotting of lysed CHO cells.

Immunohistochemisry showed highly distinct staining patterns in rat ureter and stomach.
However, western blotting of cells stably expressing the β2 receptor in CHO cells showed no difference in signal compared to lysates of cells with no endogenous expression. Thus we conclude that this antibody has no specificity for the target antigen.
