Endicott et al., BMC Res Notes describe the validation of twenty-three commercial antibodies raised against Collagen. This commonly used antibody was one of three antibodies tested that stain skeletal muscle (1:100). It also stains intracellular structures. The antibody also recognizes the target by western blot under reducing conditions (1:100).
NOTE: In the course of these studies we became aware that this mouse monoclonal antibody gave different results depending on supplier and batch number (fig 5). As shown in fig 5C, 3C4 from Santa Cruz recognized α3 at the correct molecular weight when resolved under reducing conditions. In contrast, the same antibody clone from Millipore recognized α3 at 260Kd and additional bands at 50, 70 and 90 kDa depending on lot number (fig 5.A, B). 3C4 is widely used is studies on human muscle using immunohistochemistry. Our data suggest that care should be taken in interpreting data using these batches of antibody from Millipore. See full description in the link to the article above.
