Goat Anti-ADAM17 / TACE Antibody (EB05382) from Everest Biotech.

This antibody was tested on lysate from adult mouse hippocampus and liver (picture). 50 ug protein was loaded in each lane. manufacturer suggest concentration between 0.1 to 0.3 ug/ml, so i used a concentration of 0.2ug/ml for this experiment. The antibody was left over night at 4°C in a blocking buffer. The secondary antibody was left on for 1 hour at room temperature.
A clear band was observed in the hippocampus-sample while nothing was seen in the liver, as expected. However, an unspecific band was also seen in the hippocampus sample (lower band).
