We used this antibody for IHC-labeling of human breast cancer tissue. The antibody gave a specific labeling of classical tight juntion “punctae” in ducts.
The antibody was used at 1:400 ON. The tissue was PFA-fixed, embedded in paraffin, cut at 3 um, and target retrival was performed in TEG-buffer (Tris/EGTA). The secondary antibody was used at 1:200 and developed with DAB for 1 min 15 sec.

Antibody working nice at indicated dilution. Tested in primary porcine brain endothelial cells. For IF as a secondary antibody Life technologies goat anti-mouse Alexa Fluor 568 (1:350) was used. Cells fixed with 4% PFA, permeabilized with 0.1% Triton-X100, blocked with 2% BSA.
