FLAG, Sigma-Aldrich, F1804
What do reviewers generally think about F1804 from Sigma-Aldrich?
Researchers generally report that FLAG, Sigma-Aldrich, F1804 performs well for detecting FLAG-tagged proteins in Western blot, immunofluorescence, ELISA, ChIP, and IP, often with low background and good specificity. Reported useful conditions include 1:500 for immunostaining, 1:5000 or 1:4000 for Western blot, 1:1000 for WB/IF, 0.2 µg/mL for ELISA, and 2 µg for ChIP. One reviewer points out that, although it detects the FLAG epitope, high background made it unusable in their hands.

Does detect the flag epitope, but such high background as to be unusable.

I have used this antibody for many years to verify the expression and purity of flag-tagged proteins. Never had problems of background.

I have used this antibody in 293 cells transfected with a protein containing a FLAG-tag in the N-terminus. It works great in both WB and IF (dilution 1:1000)

anti-FLAG® M2 mouse mAb Sigma-Aldrich F1804
I have used anti-FLAG in western blotting to detect bispecific diabodies (along with anti-myc), and it works perfectly. Low background and very specific detection of FLAG taged antibodies was observed. Furthermore it was possible to detect a difference in expression levels of the two chains of the diabody.
Used as 1:4000 [0.25ug/mL].

I have used this in ELISA and have had great results, very low background and high specificity. Used in concentration of 0.2ug/mL.

I have used this with great succes for quantifying expression of FLAG-tagged Rab27B using Western blot. Also, I have looked upon binding partners to FLAG-tagged Rab27B through IP.
