I am sharing a pAbmAbs generated literature review of KO validated antibodies. Here citing Huang et al. Cell & Bioscience 2021.
Huang et al. validate this antibody for detection of mouse Stat3 by both Western blotting of protein from P0 forelimbs. Western blot analysis revealed significant decrease of Stat3 protein expression in Stat3 CKO compared with control, but STAT3 band is not completely gone. There’s a faint residual band still visible (figure 1b).
Furthermore Huang et al. validate this antibody for detection of mouse Stat3 by Immunohistochemistry of bone tissues (ulna section of P0 mice). IHC staining showed the loss of Stat3 expression in Stat3 CKO (figure 1c).
Based on the available data, we have assigned a four-star rating to the Western blot and a five-star rating to IHC, but we highly encourage other scientists to share their observations.
