Researchers report that Nestin AF2736 performs well for immunofluorescence and Western blotting on RN22 cells. For immunofluorescence, the best dilution was 1:40 after brief PFA fixation and Triton X-100 permeabilization, producing a very strong signal. One reviewer points out that further dilutions may be needed for co-immunofluorescence to reduce signal. For Western blotting, a 1:1000 dilution detected two Nestin bands at approximately 250 and 220 kDa. No complaints were mentioned.

Used for Immunofluorescence of transfected RN22 cells treated with 4% PFA in PBS for 5 min. Washed 10 min in PBS. And penetrated with 0.5 % Triton x-100 in PBS for 7 min at 4 C.
Best dilution was 1:40.
Very strong signal. Try further dilutions for lesser signal, for Co-IF.
