Plectin, Abcam, ab32528

Our team has carried out deep research in the published scientific literature to identify papers and reports using antibodies directed against human Plectin and followed up by extraction and evaluation of the available data to assess its performance according to pAbmAbs rating criteria.

YCharOS is here conducting a large head-to-head study comparing 8 commercial research antibodies against human Plectin encoded by PLEC.

The collection of antibodies was tested using U2OS WT and KLEC KO cells in the applications WB (WT and KO cell lysates/10 µg protein), IP (WT cell lysate) and IF (WT and KO cells) according to YCharOS standard protocols (see reference).

In WB (1:5000 Ab dilution) ab32528 detected a band of appr. 520 and 530 kDa in WT cell lysate with no corresponding band observed in KO cell lysate. No band was detected following IP (1 µg Ab used). In IF (1:1000 Ab dilution), comparable staining intensities were observed in both KO and WT cells.

We highly encourage other scientists to share their observations.

Used for Immunofluorescence of transfected RN22 cells treated with 4% PFA in PBS for 5 min. Best dilution was 1:10, because of the low signal.

Used for Western blotting of RN22 cells in a dilution 1:5000, give one Plectin band at approximate 250 kDa.
