Phospho-Akt (Ser473), Cell Signaling Technology, 3787
What do reviewers generally think about 3787 from Cell Signaling Technology?
Researchers using Phospho-Akt (Ser473), CST 3787 at 1:50 in cultured cells report poor detection of stimulus-induced Akt phosphorylation. One reviewer points out that in hippocampal neurons stimulated with 1 nM BDNF for 10 minutes, the antibody did not detect differences in AKT phosphorylation. Another found weak staining in insulin-stimulated 293 cells across 0 to 100 nM, with somewhat more surface-associated staining at 1 and 10 nM than unstimulated controls. Overall, the reviews suggest limited sensitivity or signal for phospho-Akt changes in these applications.

I have also tested this on cultured hippocampal neurons stimulated with 1 nM of BDNF for 10 minutes. This does not detect differences in phosphorylation of AKT upon BDNF stimulation.

This antibody was tested on 293 cells in culture. The cells were stimulated with either 0, 1, 10 or 100nM of insulin before being fixed and stained. This antibody showed weak staining of cells at all concentrations. However at 1nM and 10nM the staining seemed to localised at a higher degree to the surface compared to unstimulated.
