SorCS1, VWR International, cy3
What do reviewers generally think about cy3 from VWR International?
Researchers report mixed performance for SorCS1, VWR International, cy3. One reviewer reports no Western blot signal for 510 and 511 at 1:10 in mouse adult brain, p0 brain, and CHO cells expressing mouse or human SorCS1. Another finds 511 gives specific membrane staining in CHO cells expressing mouse SorCS1 variant c, but untransfected controls show background at 1:100, with best signal/background in the second bleed. One reviewer points out that a custom rabbit second bleed detects mouse SorCS1 well in Western blots and primary cultures, without human cross-reactivity.

I have tested both 510 and 511 in Western blot. No signal at all, even when used in 1:10 dilution.
Samples: Mouse adult brain WT and SorCS1-/-, p0 WT brain and CHO cells stable expressing either mouse or human SorCS1 variant c.

Animal 511:
This antibody was used to stain CHO cells stable expressing mouse SorCS1 variant c. It stains the membrane specifically, but background staining is also seen in untransfected control cells at 1:100 concentration. The signal/background ratio is best in the second bleed.

We had this antibody custom made by Genovac using DNA immunization of rabbits with a plasmid encoding mouse SorCS1.
Second bleed is excellent in specifically detecting mouse SorCS1 in Western blotting. No cross reactivity with human SorCS1.
It also stains primary cultures derived from wild-type mice but not from SorCS1 knockouts.
