Sortilin, R&D Systems, AF3154
What do reviewers generally think about AF3154 from R&D Systems?
Researchers generally report that Sortilin, R&D Systems, AF3154 performs well for detecting sortilin in Western blotting and immunolabeling/immunostaining. One reviewer points out that it targets the extracellular domain and showed no band or labeling in sortilin knockout mouse brain, supporting specificity. It is reported to detect both human and murine sortilin in IHC at 1:100 and works well for immunostaining human hippocampal tissue at 1:250. A complaint is that it cannot be used for immunoprecipitation of sortilin from HEK293 cells after DSP crosslinking.

Review of: Sortilin, R&D Systems, AF3154 by Astrid Francisca Jacobsen
I am sharing a pAbmAbs generated literature review of KO validated antibodies. Here citing Hu et al. Frontiers in Neuroanatomy, 2017.
Hu et al. validate this antibody for detection of mice sortilin by Western blotting and immunolabeling of mouse brain lysate and tissue, respectively. This antibody targets the extracellular domain in sortilin from +/+ and -/- brains. No band was observed in the -/- mouse brain WB, and no labeling was seen in the -/- (KO mice) immunolabeling (figure 1B-C).
Based on the available data, we have assigned a five-star rating in both the Western blot and the immunolabeling, but we highly encourage other scientists to share their observations.
