pplication: I have used it for Western blotting (1:500) on wild type and Sorcs1 KO tissues (brain, kidney, muscle and liver). Brain lysate was selected as a tissue with the highest Sorcs1 expression and liver as a negative control since Sorcs1 is not expressed in the liver.
What we observe: First, we see several nonspecific bands specifically a very prominent band at around 75kDa. This band does not represent SORCS1 as the molecular weight of SORCS1 is around 130kDa and not 75kDa. Second, we see a band around the expected molecular weight for SORCS1 but this band is also present in both the WT and KO samples. Third, there is no change in band appearance or intensity between brain and liver lysate – tissue with the highest and no expression of SORCS1.
Conclusion: This is not a good/specific antibody for SORCS1.

Detects both mouse and human SorCS1 in Wetsern blots for samples of CHO cells stable expressing each variant of SorCS1.
On the other hand, it does not work for detecting endogenous murine SorCS1 from brains of p0 and Adult mice in Western blotting. A SorCS1 knockout brain were used as negative reference. Cells expressing SorCS1 as positive ref.

This antibody works in western blotting of HEK cells expressing human SorCS1(c).
It also works on tissues subjected to human SorCS1 virus. Please see the image above. Lane 4 is lysate of untransfeceted cells.

This antibody was used to immunostain CHO cells stable expressing mouse SorCS1 variant c. It stains specific and with low background in untransfected cells at a 1:100 concentration.
