Researchers generally report that TrkB AF1494 performs well for mouse TrkB detection by Western blot, immunocytochemistry, immunoprecipitation, and IHC, often giving clear full-length and T1 splice-variant bands. This antibody works very well for detection of mouse TrkB. Clear bands of both full-length and splice variant T1 are visible using 1:1000 dilution.

Works very well for ICC of primary hippocampal neurons.
Neurons were fixed for 20 min in 4% PFA and permeabilized using PBS + 0.1% Triton X-100. After blocking, the neurons were incubated with primary antibody in a 1:500 dilution in blocking buffer.

Mouse TrkB antibody, AF 1494, R&D Systems
I have used this antibody for western blotting of call lysates from a primary hippocamal neuron culture in a dilution of 1:1000. This antibody works so well for me, that I often use it as a control of my blotting protocol.

Also works extremely well with immunocytochemistry of human TrkB. i aplied it in a 2 ug/ml and got a clearcut window with no control-staining

I have used this antibody for IHC on hippocampaltissue from 10 days old mice. A clear staining was observed which was not seen in the negative control.

I have used this antibody for western blot and IHC on mouse olfactory bulb and mouse hippocampus. This antibody is extremely good for western blot. But IHC staining for hippocampus did not work. Also I performed IHC on spinal cord cryosections. It works very good with the right protocol. I recommend to follow the exact protocol for IHC in R&D website.
For western, the dilution is: 1/1000
For IHC on spinal cord cryosections, the dilution is: 1/500

I have tested this antibody for IP of human TrkB. Although it works very well for IP of mouse TrkB, it does not work for IP of human TrkB.

I have tested this antibody for IP as well as western blot on cultured hippocampal neurons with very good results. Similar to Simon Glerups review i detected two bands corresponding to full lenght and splice variant.
IP and WB i used 0.2 ug/ml

This antibody is excellent!
We went through 15 different commercial antibodies before finding it.
It works for IP of endogenous TrkB from hippocampal neurons, for ICC and IHC on neurons and cryosections, and for Western blotting. With this antibody you will achieve superior sensitivity when doing Western blotting of tissues and blots can be stripped and reprobed several times and still give a nice clean specific band.
The Western blot above shows full length TrkB and the splice variant TrkB T1 in synaptosomal preparations of mouse hippocampus.
For WB we use 0.2 ug/ml.
For ICC we use 2 ug/ml
