I am sharing a pAbmAbs-generated literature review of KO-validated antibodies, citing Gunesch et al., eLife (2020). Gunesch et al. validated this antibody for the detection of endogenous human CD56 (NCAM1) by Western blotting using WT and CD56-KO YTS and NK92 cell lysates. CD56-KO cells were generated using CRISPR/Cas9. In WT cells, CD56 was detected over a molecular-weight range of approximately 130–160 kDa, consistent with differentially polysialylated forms of CD56. No corresponding CD56 signal was detected in the CD56-KO YTS or NK92 cell lysates (Figure 1A). No full-length/uncropped blot images were available, limiting assessment of potential off-target bands outside the displayed molecular-weight range. Based on the available data, we have assigned this antibody a four-star rating. However, we strongly encourage other scientists to share their observations and experiences with this antibody.
