I am sharing a pAbmAbs-generated literature review of KO-validated antibodies, citing Mikedis et al., eLife (2020). Mikedis et al. evaluated this antibody for the detection of endogenous mouse DAZL by immunofluorescence staining of testis sections from control and conditional Dazl-KO mice. The antibody was used at a dilution of 1:100. The conditional deletion of Dazl was incomplete, resulting in mosaic cKO seminiferous tubules containing both DAZL-positive and DAZL-negative germ cells. Clear DAZL staining was observed in control cells and in residual DAZL-positive cells within mosaic cKO tubules, whereas DAZL staining was absent from DAZL-negative germ cells and DAZL-negative cKO tubules (Figure 1C). Based on the available data, we have assigned this antibody a four-star rating. The incomplete and mosaic nature of the conditional knockout limits direct comparison between control and KO tissue; however, the absence of staining in genetically DAZL-negative cells provides evidence supporting antibody specificity. We strongly encourage other scientists to share their observations and experiences with this antibody.
