I am sharing a pAbmAbs-generated literature review of KO-validated antibodies, citing Schade et al., Oncogene (2019). Schade et al. validated this antibody for the detection of endogenous human RB1 by Western blotting using WT and RB1-KO cells. RB1 knockout was generated using CRISPR/Cas9 and validated in multiple human cell lines, including HFF, A549, and T47D cells. A clear RB1 band was detected in control cells, whereas the corresponding band was absent in RB1-KO cells (Figures 4A, 7B, and 7F). In Figure 7F, an additional nonspecific band was also observed. Based on the available data, we have assigned this antibody a five-star rating. The antibody shows clear loss of the expected RB1 signal upon knockout across multiple independent human cell lines. However, we strongly encourage other scientists to share their observations and experiences with this antibody.
