Review of: ApoE, Abcam, ab183597 by pAbmAbs data mining
We generated a top 1000 list of the most studied and most published proteins and analyzed literature validation data for commercially available research antibodies. APOE encoding apolipoprotein E (apoE) is the 5th most studied gene on the list.
In Ferguson et al., Alzheimer’s & Dementia (2024), the authors investigate APOE silencing in mouse models of Alzheimer’s disease. Abcam ab183597 (EPR19392) is tested by capillary WB of homogenates of hippocampus, cortex and liver samples from individual mice treated with APOE-targeting or non-targeting siRNA. Images are shown in Supplementary Figure S2, with quantification in Figure 1D and Supplementary Figure S1D.
ApoE signal at 37 kDa becomes undetectable in hippocampus and cortex and is reduced in liver after brain-directed siRNA. Liver-directed siRNA eliminates detectable liver signal while retaining brain signal. Vinculin provides a loading control, and KD is supported by mRNA measurements. These data strongly support specific APOE detection within the displayed MW range.
We have rated ab183597 under the WB application based on capillary WB data and according to the pAbmAbs criteria.
