I am sharing a pAbmAbs generated literature review of KO validated antibodies. Here citing Lubbers et al. JBC, 2019. Lubbers et al. validated this antibody for the detection of mouse αII-spectrin (SPTAN1) by Western blotting and immunofluorescence using isolated adult ventricular cardiomyocytes from control and cardiomyocyte-specific αII-spectrin knockout (cKO) mice. The cKO mice were generated using the Cre-lox system. By WB, a strong reduction in both full-length αII-spectrin (>250 kDa) and a ~150 kDa degradation product was observed in cKO cardiomyocytes compared with control cells (Figure 2B). No off-target bands can be assessed outside the displayed molecular-weight region because only cropped Western blot images were available Immunofluorescence staining showed clear αII-spectrin staining in control cardiomyocytes (Figure 2H), whereas αII-spectrin staining was absent in cKO cardiomyocytes (Figure 2J). Based on the available data, we have assigned a four-star rating for WB and a five-star rating for IF, but we highly encourage other scientists to share their observations.
