I am sharing a pAbmAbs-generated literature review of KO-validated antibodies, citing Yang et al., Frontiers (2019). Yang et al. validated this antibody for the detection of human SREBP1 by Western blotting of WT and SREBP1-KO HCC cell lysates. The KO cells were generated using the lentiCRISPRv2 system. No detectable band was observed in the SREBP1-KO cells, whereas a strong band was present in the WT cell lysate (Figure 1C). However, only a cropped Western blot image was provided, preventing assessment of potential off-target bands or overall antibody specificity across the full molecular-weight range. Based on the available data, we have assigned this antibody a four-star rating, but we highly encourage other scientists to share their observations.
