Review of: Ubiquitin-specific protease 30, Cell Signaling Technology, 57045 by pAbmAbs Data Mining
Our team has carried out deep research in the published scientific literature to identify papers and reports using antibodies directed against human Ubiquitin-specific protease 30 (USP30) and followed up by extraction and evaluation of the available data to assess its performance according to pAbmAbs rating criteria.
YCharOS is here conducting a large head-to-head study comparing 7 commercial research antibodies against human Ubiquitin-specific protease 30 (USP30).
The collection of antibodies was tested using MCF-7 and HAP1 (WT and USP30 KO cells) in the application WB (WT and KO cell lysates/30 µg protein), IP (MCF-7 WT cell lysate) and IF (MCF-7 WT and KO cells) according to YCharOS standard protocols (see reference).
In WB (1:1000 ab dilution) 57045 detected a band of appr. 58 in WT cell lysate with no corresponding band observed in KO cell lysate. This pattern was consistent in both HAP1 and MCF-7 cell lysate. A band was detected following IP (10 µl Ab used), this band was markedly more intense compared to band in input extract (6% of starting material) and no band was detected in immunodepleted extract (6% of total unbound fraction), showing that the antibody does completely deplete the antigen from cell lysate. In IF (1:100 ab dilution), clear intense staining was observed only in WT cells.
