Review of: Guanine nucleotide-binding protein G(q) subunit alpha, Cell Signalling Technology, 14373 by pAbmAbs Data Mining
Our team has carried out deep research in the published scientific literature to identify papers and reports using antibodies directed against human Guanine nucleotide-binding protein G(q) subunit alpha (GNAQ) and followed up by extraction and evaluation of the available data to assess its performance according to pAbmAbs rating criteria.
YCharOS is here conducting a large head-to-head study comparing 11 commercial research antibodies against human Guanine nucleotide-binding protein G(q) subunit alpha (GNAQ)
The collection of antibodies was tested using HAP1 WT and GNAQ KO cells in the applications WB (WT and KO cell lysates/30 µg protein) and flow cytometry (WT and KO cells) according to YCharOS standard protocols (see reference).
In WB (1:1000 ab dilution) 14373 detected a band of appr. 42 kDa in WT cell lysate with no corresponding band observed in KO cell lysate. Furthermore, faint bands of higher molecular weight were observed in both genotypes. Following flow cytometry (1:590 ab dilution) WT cells displayed increased fluorescence intensity compared with knockout cells, consistent with target-dependent binding. However, knockout cells retained a substantial signal above the secondary-antibody control, indicating significant background staining. These results support partial but not complete specificity of the antibody for the target antigen.
