Review of: C-Myc, Abcam, ab32072 by Astrid Francisca Jacobsen
I am sharing a pAbmAbs-generated literature review of KO-validated antibodies, citing Littler et al., Open Biology(2019).
Littler et al. validated this antibody for detection of human c-MYC by Western blotting in RKO cells. In FC-MYC cells, the endogenous MYC alleles were disrupted by CRISPR/Cas9, resulting in a strong reduction of the MYC signal in the absence of tetracycline. MYC expression, and the corresponding antibody signal, was restored upon induction of the tetracycline-regulated MYC transgene (Figure 1b). The antibody was also evaluated by immunostaining, where MYC staining was strongly reduced in the MYC-deficient cells and restored following tetracycline-induced MYC expression (Figure 1c).
Based on the available data, we assign a four-star rating for Western blotting. The genetically controlled loss and restoration of MYC signal provide strong evidence for target-specific detection; however, the absence of a reported antibody dilution and full-range Western blot data limits reproducibility and assessment of potential off-target reactivity. We assign a five-star rating for immunofluorescence. We highly encourage other scientists to share their observations.
