I am sharing a pAbmAbs generated literature review of KO validated antibodies. Here citing Abe et al. Molecular and cellular Biology, 2019. Abe et al. validate this antibody for detection of human IKKa by Western blotting of WT and IKKa KO 293T cell lysates. KO cells were transfected with expression constructs for HA-tagged TLR3 and Flag-tagged IRF-3 S386A and stimulated with 50 μg/ml of poly(I·C) for 2 h. No band was observed in IKKa KO 293T cells, compared to WT cell lysate were a band of appr. 85 kDa (Figure 4G). However, only the cropped image were available, why potential off-target bands outside the displayed molecular-weight range cannot be assessed. Based on the available data, we have assigned a four-star rating, but we highly encourage other scientists to share their observations.
