NOXA, Sigma-Aldrich, OP180

pAbmAbs reviewed Bajpai et al. Nature Communications (2020), in which authors evaluated Sigma-Aldrich OP180 against NOXA in Western blotting. The antibody was tested using RPMI-8226 and KMS18 multiple myeloma cells and validated using genetic validation, including CRISPR/Cas9-generated NOXA knockout cells (Figure 5l and Figure 5n). In Western blotting (1:1000), the antibody detected NOXA in control cell lysate in both cell lines, however residual bands were detected in KMS18 NOXA KO cell lysate (Figure 5l), whereas no band was observed in RPMI-8226 NOXA KO cell lysate (Figure 5n). Only cropped Western blot regions are presented for 5l, so potential off-target bands outside the displayed molecular-weight range cannot be assessed. Based on the available experimental evidence and pAbmAbs review criteria, we assign 4 stars for Western blotting. The partial loss of NOXA signal in two independent CRISPR/Cas9 NOXA-knockout cell lines strongly support target-specific detection by OP180, but the absence of uncropped/full-range Western blot data prevents assessment of potential off-target reactivity across the full molecular-weight range.
