siRNA KD of TGFB1 in human cancer cell lines

We generated a top 1000 list of the most studied and most published proteins and analyzed literature validation data for commercially available research antibodies. TGFB1 encoding Transforming Growth Factor Beta 1 (TGF-beta-1) is the 7th most studied gene on the list. In Wang et al., NAR Cancer; 8 (2026), the authors investigate TGF-β1 silencing in cancer therapy. Abcam ab215715 is used for WB of human PANC-1 and MDA-MB-231 cell lysates following siRNA TGFB1 KD, compared with untreated and non-targeting siRNA control (protein amount not reported). Data are shown in Figure 3A. The detected band decreases with increasing siRNA concentration in both cell lines, supporting specific detection of TGF-β1 by ab215715 in WB under the tested conditions. The authors describe total-protein normalization, but no loading control is shown. The blot is cropped and lacks molecular weight markers, limiting assessment of band size, sample loading and nonspecific bands. Abcam ab215715 (1:1000) is used for WB of rat bone marrow-derived endothelial progenitor cell lysates following TGFB1 knockdown with three different siRNAs, compared with a non-targeting siRNA control (protein amount not reported). Data are shown in Figure 4D. The detected band is reduced with two of the three TGFB1 siRNAs. GAPDH provides a loading control. This supports specific detection of TGF-β1 by ab215715 in WB under the tested conditions. However, the blot is cropped and lacks molecular weight markers, limiting assessment of band size and nonspecific bands.
