AT1R, GeneTex, GTX89149

pAbmAbs reviewed the publication entitled “Validation of six commercially available angiotensin II type 1 receptor antibodies”, in which Bingjie Li et al. evaluated GeneTex GTX89149 against the angiotensin II (Ang II) type 1 receptor (AT1R) in WB and IF. The antibody was tested using cardiomyocyte conditional knockout C57BL/6N mice, human AT1R-overexpressing CHO stable-transformed cell lines and human AT1R-overexpressing HEK293 cells. AT1R expression and localization were assessed through receptor-ligand binding assays, RT-PCR, WB, and IF. The WB analysis of GTX89149 is presented in Figure 1. Only very narrowly cropped blots are shown. Uncropped blots are not available. The signal from GTX89149 is unchanged in knockouts and up upon overexpression. Thus, based on the available data and our rating criteria, no more than a 1-star rating can be granted. The IF analysis of GTX89149 is presented in Figure 2. No specific AT1R signal could be detected in overexpressing CHO cells compared to untransfected CHO cells. Thus a 1-star rating is granted. We encourage other scientists to share their observations.
