AT1R, Thermo Fisher Scientific, PA5-18587

pAbmAbs reviewed the publication entitled “Validation of six commercially available angiotensin II type 1 receptor antibodies”, in which Bingjie Li et al. evaluated ThermoFisher PA5-18587 against the angiotensin II (Ang II) type 1 receptor (AT1R) in WB and IF. The antibody was tested using cardiomyocyte conditional knockout C57BL/6N mice, human AT1R-overexpressing CHO stable-transformed cell lines and human AT1R-overexpressing HEK293 cells. AT1R expression and localization were assessed through receptor-ligand binding assays, RT-PCR, WB, and IF. The WB analysis of PA5-18587 is presented in Figure 1. Only very narrowly cropped blots are shown. Uncropped blots are not available. The signal from PA5-18587 is unchanged in knockouts and up upon overexpression. Thus, based on the available data and our rating criteria, no more than a 1-star rating can be granted. The IF analysis of PA5-18587 is presented in Figure 2. No specific AT1R signal could be detected in overexpressing CHO cells compared to untransfected CHO cells. Thus a 1-star rating is granted. We encourage other scientists to share their observations.
