AT1R, Abclonal, A14201

pAbmAbs reviewed the publication entitled “Validation of six commercially available angiotensin II type 1 receptor antibodies”, in which Bingjie Li et al. evaluated ABclonal A14201 against the angiotensin II (Ang II) type 1 receptor (AT1R) in WB and IF. The antibody was tested using AT1R global knockout SD rats, cardiomyocyte conditional knockout C57BL/6N mice, human AT1R-overexpressing CHO stable-transformed cell lines and human AT1R-overexpressing HEK293 cells. AT1R expression and localization were assessed through receptor-ligand binding assays, RT-PCR, WB, and IF. The WB analysis of A14201 is presented in Figure 1. Only very narrowly cropped blots are shown. Uncropped blots are not available. There is some indication that the signal from A14201 goes down in knockouts and up upon overexpression. However, this is not convincing, and based on the available data and our rating criteria, no more than a 2-star rating can be granted. The IF analysis of A14201 is presented in Figure 2. No specific AT1R signal could be detected in WT cardiomyocytes vs KO or in overexpressing CHO cells. Thus a 1-star rating is granted. We encourage other scientists to share their observations.
