Review of: MTHFR, GeneTex, GTX100535 by pAbmAbs data mining
We generated a top 1000 list of the most studied and most published proteins and analyzed literature validation data for commercially available research antibodies. MTHFR encoding methylenetetrahydrofolate reductase is the eighth most studied gene on the list.
In Tong et al., Oncogenesis (2020), the authors investigate the regulation of gastric cancer cell proliferation, including the contribution of MTHFR. GTX100535 is used for WB of AGS and MKN45 lysates from cells treated with MTHFR siRNAs. The data is shown in Figure 4E.
GTX100535 detects a band labelled 72 kDa, consistent with the expected size of MTHFR. The antibody signal is substantially reduced compared to control cells. This is observed with two independent siRNAs in both cell lines, β-actin loading controls shown. Data supports specific detection of MTHFR in WB under the tested conditions. The blots are cropped, limiting assessment of additional nonspecific bands.
