Review of: MTHFR, Abcam, ab203786 by pAbmAbs data mining
We generated a top 1000 list of the most studied and most published proteins and analyzed literature validation data for commercially available research antibodies. MTHFR encoding methylenetetrahydrofolate reductase is the eighth most studied gene on the list.
In Su et al., Cancer Discovery (2020), the authors investigate how MTHFR affects the response of acute myeloid leukemia cells to MYC-targeting therapies. Ab203786 is used for WB of mouse bone-marrow-derived leukemia cells derived from Mthfr WT, heterozygous and KO animals. The data are shown in Figure 4G.
The antibody detects a band in WT samples, with weaker signals in heterozygotes and no detectable corresponding band in KO samples. Vinculin provides a loading control. Additional shRNA KD comparisons in human and mouse leukemia cells support band specificity (Figures 2B and 2G). Together, data support specific detection of MTHFR by ab203786 in WB under the tested conditions. The blots are cropped and lack molecular-weight markers, limiting assessment of band size and additional nonspecific bands.

In Young et al., Zygote (2022), the authors investigate MTHFR phosphorylation during mouse oocyte maturation. Abcam ab203786 was used for WB (1:1000) of liver homogenates (5 µg) and oocyte lysates obtained from WT and Mthfr KO mice. The data are shown in Figure 3A–B and E, with complete blot scans provided in Figures S1–S3.
The antibody detects bands near 75 kDa in WT liver, with weaker signals in heterozygotes and no corresponding bands in KO samples. The oocyte signal is also absent in KO samples. Vinculin and GAPDH provide loading controls. Data support specific detection of MTHFR by ab203786 in WB under the tested conditions. The authors report a nonspecific band below 50 kDa in liver lysates.
