We generated a top 1000 list of the most studied and most published proteins and analyzed literature validation data for commercially available research antibodies. ESR1 encoding estrogen receptor alpha (ERα) is the ninth most studied gene on the list. In Sullivan et al., Journal of Applied Physiology (2024), the authors use skeletal muscle-specific Esr1 KO mice to investigate the role of ERα in muscle contractility. Abcam ab32063 (1:5000) is used for WB of skeletal muscle lysates (30 µg) from male KO mice and WT littermate controls. The data are shown in Figure 1B. The antibody detects a faint band near the expected molecular weight of 66 kDa in three WT samples with no corresponding band apparent in KO samples. Uterus lysate provides a strong positive control. These data support specific detection of ERα by ab32063 in WB under the tested conditions. However, the weak muscle signal, cropped blot and absence of a loading control limit assessment of sample loading and additional nonspecific bands. We have rated ab32063 based on the available information and according to the pAbmAbs criteria. We highly encourage other scientists to share their observations.
