NFκB p50, Santa Cruz Biotechnology, sc-8414

We generated a top 1000 list of the most studied and most published proteins and analyzed literature validation data for commercially available research antibodies. NFKB1 is the seventh most studied gene on the list. In Herkenham et al., Journal of Neuroinflammation 8:141 (2011), the authors tested antibodies targeting NFKB1 p50 by Western blot using tissue homogenates from wild-type and Nfkb1 knockout mice using 50 µg of mouse brain or spleen homogenate per lane. The data for sc-8414 ia shown in Figure 3d of the paper. In spleen homogenate, sc-8414 detects a prominent band just above the expected size of p50. This band is markedly reduced in Nfkb1 knockout tissue. A fainter band slightly above the expected p50 position is also visible in wild-type brain but not apparent in the knockout sample. However, the blots contain numerous additional bands in both tissue homogenates, including prominent higher molecular weight bands that are also detected in knockout tissues. These results suggest detection of NFKB1 specifict bands, alongside substantial nonspecific binding under the tested WB conditions. We have rated sc-8414 based on the available information and according to the pAbmAbs criteria. We highly encourage other scientists to share their observations.
