I am sharing a pAbmAbs-generated literature review of KO-validated antibodies, citing Sola et al., Communications Biology (2020). Sola et al. validated this antibody for the detection of human Tau by Western blotting and immunostaining in SH-SY5Y neuroblastoma cells. KO cells were generated using CRISPR/Cas9, and the 231K cell line showed successful deletion of the Tau gene. In Western blotting, only a faint band was detected in the KO cell lysate, whereas an intense band accompanied by some smearing was observed in the WT cell lysate (Figure 1A). Immunostaining showed only a few speckles in the KO cells, while intense staining was observed in the WT cells (Figure 1A). Based on the available data, we have assigned this antibody a four-star rating for Western blotting and a five-star rating for immunofluorescence. However, we strongly encourage other scientists to share their observations and experiences with this antibody.
