Review of: EGFR (E746-A750del Specific), Cell Signaling Technology, 2085 by pAbmAbs Data Mining

Cell Signaling 2085 was validated in the original peer-reviewed paper from Cell Signaling Technology by Yu et al., 2009, Clinical Cancer Research entitled Mutation-Specific Antibodies for the Detection of EGFR Mutations in Non–Small-Cell Lung Cancer
The authors tested the mutation-specific antibody by Western blot, immunofluorescence, and immunohistochemistry.
Clone 6B6/#2085 was screened against six human cancer cell lines with known EGFR genotypes. It detected E746_A750del EGFR in H1650 and HCC827, but not WT EGFR in Kyse450/Kyse70 or the L858R mutant recognized by the other mutation-specific antibody described in the publication.
The #2085 antibody was tested on paraffin-embedded NSCLC specimens and compared against independently determined EGFR mutation status by DNA sequencing. Across the two mutation-specific antibodies, the IHC assay achieved 92% sensitivity and 99% specificity relative to sequencing. Yu et al. report that the antibody specifically detected their corresponding mutant EGFR forms by immunofluorescence as well.
