I am sharing a pAbmAbs generated literature review of KO validated antibodies. Here citing Dilshat et al. Cancer Biology, Genetics and Genomics 2021. Dilshat et al. evaluated this antibody for the detection of endogenous human MITF by Western blotting (WB) and immunofluorescence (IF) using control and MITF-KO SkMel28 cells. In WB (1:2,000 antibody dilution), ab12039 detected multiple bands in EV-SkMel28 control cell lysates. A prominent band at approximately 60 kDa was observed, along with additional bands at approximately 40 and 75 kDa. Fainter bands at corresponding molecular weights were observed predominantly in MITF-X2 and, to a lesser extent, in MITF-X6 (Figure 1D). In IF (1:200 antibody dilution), staining was observed in both EV-SkMel28 and MITF-X2 cells, whereas MITF-X6 cells exhibited weaker staining (Figure 1E). Based on the available data, we have assigned a two-star rating for both WB and IF but we highly encourage other scientists to share their observations.
