I am sharing a pAbmAbs generated literature review of KO validated antibodies. Here citing Cui et al. Nature Communications 2021. Cui et al. validated this antibody for the detection of endogenous human FTO by Western blot analysis of wild-type (WT) As-T cell lysates and CRISPR-generated FTO knockout (KO) cell lysates derived from two independent sgRNAs. No band was observed in FTO KO-1 or FTO KO-2 As-T cell lysate (Figure 2a). Based on the available data, we have assigned a five-star rating. However, as the presented blot is cropped, this assessment should be interpreted with some caution. We strongly encourage other researchers to share their observations and validation data.
