Review of: ARID1A, GeneTex, GTX638371 by pAbmAbs Data Mining
Our team has carried out deep research in the published scientific literature to identify papers and reports using antibodies directed against human AT-rich interactive domain-containing protein 1A (ARID1A) and followed up by extraction and evaluation of the available data to assess its performance according to pAbmAbs rating criteria.
YCharOS is here conducting a large head-to-head study comparing 14 commercial research antibodies against human AT-rich interactive domain-containing protein 1A (ARID1A).
The collection of antibodies was tested using HAP1 WT and ARID1A KO cells in the applications WB (WT and KO cell lysates/30 µg protein), IP (WT cell lysate) and IF (WT and KO cells) according to YCharOS standard protocols (see reference).
In WB (1:500 ab dilution) using GTX638371, multiple bands are detected in both WT and knockout samples, including bands within the expected molecular weight range of the target protein (~245 kDa). As these signals are not abolished in the knockout sample, the specificity of the antibody for the target protein cannot be confirmed. A band was detected following IP (2 µg Ab used), this band was markedly more intense compared to band in input extract (6% of starting material) and no band was detected in immunodepleted extract (6% of total unbound fraction), showing that the antibody does completely deplete the antigen from cell lysate. In IF (1:1000 ab dilution), comparable staining was observed in KO and WT cells.
