Review of: ARID1A, GeneTex, GTX129433 by pAbmAbs Data Mining
Our team has carried out deep research in the published scientific literature to identify papers and reports using antibodies directed against human AT-rich interactive domain-containing protein 1A (ARID1A) and followed up by extraction and evaluation of the available data to assess its performance according to pAbmAbs rating criteria.
YCharOS is here conducting a large head-to-head study comparing 14 commercial research antibodies against human AT-rich interactive domain-containing protein 1A (ARID1A).
The collection of antibodies was tested using HAP1 WT and ARID1A KO cells in the applications WB (WT and KO cell lysates/30 µg protein), IP (WT cell lysate) and IF (WT and KO cells) according to YCharOS standard protocols (see reference).
In WB (1:500 ab dilution) GTX129433 detects a faint WT-specific signal at the expected high-molecular-weight region (~245 kDa), which is absent in the knockout sample. The immunoreactive signal appears as a cluster of closely migrating bands rather than a single distinct band. Furthermore, faint bands of lower molecular weight were observed in both genotypes. A band was detected following IP (2 µg Ab used), this band was more intense compared to band in input extract (6% of starting material). A band was also detected in immunodepleted extract (6% of total unbound fraction), showing that the antibody does not completely deplete the antigen from cell lysate. In IF (1:250 ab dilution), comparable staining was observed in KO and WT cells.
