Our team has carried out deep research in the published scientific literature to identify papers and reports using antibodies directed against human pyruvate carboxylase and followed up by extraction and evaluation of the available data to assess its performance according to pAbmAbs rating criteria.

YCharOS is here conducting a large head-to-head study comparing 13 commercial research antibodies against human pyruvate carboxylase encoded by PC.

The collection of antibodies was tested using HAP1 WT and PC KO cells in the applications WB (WT and KO cell lysates/30 µg protein), IP (WT cell lysate) and IF (WT and KO cells) according to YCharOS standard protocols (see reference).

In WB (1:10000 ab dilution) 16588-1-AP detected an intense band of appr. 130 kDa in WT cell lysate not observed in KO cell lysate. Bands of lower molecular weight was observed in both genotypes. A band was detected following IP (2 µg Ab used); however, this band was less intense compared to bands in input extract (6% of starting material) and immunodepleted fraction (6% unbound fraction), indicating that the antibody did not efficiently deplete the antigen from the cell lysate. In IF (1:500 ab dilution), more intense staining was observed in WT cells compared to KO cells exhibiting background staining.
