Review of: NRF2, ABclonal, A21176 by pAbmAbs Data Mining
Our team has carried out deep research in the published scientific literature to identify papers and reports using antibodies directed against human nuclear factor erythroid 2-related factor 2 (NRF2) and followed up by extraction and evaluation of the available data to assess its performance according to pAbmAbs rating criteria.
YCharOS is here conducting a large head-to-head study comparing 17 commercial research antibodies against human nuclear factor erythroid 2-related factor 2 (NRF2) encoded by NFE2L2.
The collection of antibodies was tested using HCT116 WT and NFE2L2 KO MG-132 treated cells in the applications WB (WT and KO cell lysates/30 µg protein), IP (WT cell lysate) and flow cytometry (WT and KO cells) according to YCharOS standard protocols (see reference).
In WB (1:1000 ab dilution) detected a faint band of appr. 95 kDa in WT cell lysate with no corresponding band observed in KO cell lysate. Faint bands of lower molecular weight were observed in WT cell lysate. A band of comparable intensity to the starting material (6%) was observed in the unbound fraction (6%), however the immunoprecipitated fraction was not shown. Flow cytometry using A21176 (1:11000 ab dilution) showed overlapping fluorescence intensity distributions for untreated WT and KO cells, with no visible shift between the two populations. When stimulated with MG-132 a right shift in WT cells were observed.
