Review of: CDK5, Cell Signaling Technology, 14145 by pAbmAbs Data Mining
Our team has carried out deep research in the published scientific literature to identify papers and reports using antibodies directed against human Cyclin-dependent kinase 5 and followed up by extraction and evaluation of the available data to assess its performance according to pAbmAbs rating criteria.
YCharOS is here conducting a large head-to-head study comparing 13 commercial research antibodies against human Cyclin-dependent kinase 5 encoded by CDK5.
The antibody was tested using HAP1 WT and CDK5 KO cells in Western blot (WT and KO cell lysates, 30 µg total protein) and additionally by WB in HEK293T cells (KO and WT), as well as in immunoprecipitation (IP; WT cell lysate) and immunofluorescence (IF; WT and KO cells), according to YCharOS standard protocols (see reference).
In WB (1:1000 ab dilution) 14145 detected a band of appr. 33 kDa in WT cell lysate with no corresponding band observed in KO cell lysate. This pattern was consistent in both HAP1 and HEK293T cell lysate. Immunoprecipitation using 2 μg of antibody failed to detect a putative target band. In IF (1:250 ab dilution), clear staining was observed in WT cells with no background staining observed in KO cells.
