Our team has carried out deep research in the published scientific literature to identify papers and reports using antibodies directed against human The tripartite motif containing-21(TRIM21) and followed up by extraction and evaluation of the available data to assess its performance according to pAbmAbs rating criteria.

YCharOS is here conducting a large head-to-head study comparing 10 commercial research antibodies against human The tripartite motif containing-21encoded by the TRIM21 gene.

The collection of antibodies was tested using A549 WT and TRIM21 KO cells in the applications WB (WT and KO cell lysates/30 µg protein), IP (WT cell lysate) and IF (WT and KO cells) according to YCharOS standard protocols (see reference).

In WB (1:1000 ab dilution) 67136-1-Ig detected a band og appr. 50 kDa in WT cell lysate with no corresponding band observed in KO cell lysate. A strong band of lower molecular weight was observed in KO cell lysate. Extracts from A549 WT, A549 TRIM21 KO, PANC-1, SU.86.86, H4, 5637, BxPC-3, A-172, U-87 MG and FaDu cells were processed for western blot and 67136-1-Ig detected a band in all different cell types – in PANC-1, SU.86.86, 5637, BxPC-3 and FaDu a band of lower molecular weight was observed. A band was detected following IP (2 µg Ab used), this band was more intense compared to band in input extract (5% of starting material). No band was also detected in immunodepleted extract (5% of total unbound fraction), showing that the antibody does completely deplete the antigen from cell lysate. In IF (1:800 ab dilution), KO cells showed stronger staining intensity compared to WT cells.
