Review of: STAT5B, Abcam, ab178941 by pAbmAbs Data Mining
Our team has carried out deep research in the published scientific literature to identify papers and reports using antibodies directed against human Signal transducer and activator of transcription 5B and followed up by extraction and evaluation of the available data to assess its performance according to pAbmAbs rating criteria.
YCharOS is here conducting a large head-to-head study comparing 13 commercial research antibodies against human Signal transducer and activator of transcription 5B encoded by STAT5B.
The antibody panel was tested according to YCharOS standard protocols (see reference) using HAP1 WT and VPS35 KO cells in Western blot (WB; WT and KO cell lysates, 20 µg protein), immunoprecipitation (IP; WT cell lysate), and immunofluorescence (IF; WT and KO cells). Furthermore, the antibody was also evaluated in WB using HeLa WT and STAT5B KO cell lysate.
In WB (1:1000 ab dilution) ab178941 detected a band of appr. 90 kDa in WT cell lysate with no corresponding band observed in KO cell lysate. This pattern was consistent in both HAP1 and HeLa cell lysate. A band was detected following IP (2 µg Ab used), this band was markedly more intense compared to band in input extract (10% of starting material) and no band was detected in immunodepleted extract (10% of total unbound fraction), showing that the antibody does completely deplete the antigen from cell lysate. In IF, a more intense staining was observed in WT cells compared to KO cells showing some background.
