Our team has carried out deep research in the published scientific literature to identify papers and reports using antibodies directed against human mitochondrial 2-oxoglutarate/malate carrier protein and followed up by extraction and evaluation of the available data to assess its performance according to pAbmAbs rating criteria.

YCharOS is here conducting a large head-to-head study comparing 6 commercial research antibodies against human mitochondrial 2-oxoglutarate/malate carrier protein encoded by SLC25A11.

The collection of antibodies was tested using HEK293T WT and SLC25A11 KO cells in the applications WB (WT and KO cell lysates/25 µg protein) and IP (WT cell lysate). This antibody was furthermore tested in HAP1 (WT and KO cell lysate/40 µg protein). All experiments were done according to YCharOS standard protocols (see reference).
In WB (1:1000 ab dilution) 12253-1-AP detected a band of appr. 31 kDa in WT cell lysate with no corresponding band observed in KO cell lysate. An intense band of higher molecular weight was observed in both genotypes. This pattern was consistent in both HAP1 and HEK293T cell lysate. A band was detected following IP (2 µg Ab used), this band had same intensity compared to bands in input extract (4% of starting material) and immunodepleted extract (4% of total unbound fraction), showing that the antibody does not deplete the antigen from cell lysate.
